A research team from West China Hospital of Sichuan University published a study in the MedComm journal showing that supplementing with NMN or NR can increase NAD synthesis, enhance Sirt1 activity, maintain mitochondrial function, protect nuclear DNA integrity, anti-inflammatory, promote cellular antioxidant function, thereby resisting replication and stress-induced aging and promoting lung health.

NMN alleviates aging of alveolar cells

As one ages, the function of various tissues and organs in the human body gradually declines, including the lungs. Among them, the aging of alveolar epithelial cells (ACEs) is one of the main reasons for the decline in respiratory defense and function. The aging process of the lungs is accompanied by a decrease in the number and function of alveolar epithelial cells (ACE). At the same time, the decrease in intracellular NAD is also accompanied by aging.

Researchers fed NMN to 8-10 month old C57/BL6 mice at a dose of 500 mg/kg/day for 2 months. Afterwards, the aging indicators of lung tissue were tested.

The results showed that compared with the aging group, the aging related cyclic inhibitory proteins in the gastric aging+NMN group and the water aging+NMN group were significantly reduced, indicating that long-term addition of NMN may effectively improve lung aging.

NMN alleviates cellular aging

In order to investigate whether NMN can directly alleviate the replicative and stress-induced aging of ACE, researchers conducted animal experiments.

They added NMN to the cells from the 4th passage to the 7th passage, and compared with the control group, they found a significant improvement in the aging phenotype of the cells.

Among them, aging related SA was found in the 7th/8th generation- β- Gal staining significantly decreased in ACE. In summary, the addition of NMN can effectively alleviate the replication of primary pulmonary ACEs.

NMN improves pulmonary inflammation

Bleomycin (BLM) is a member of the glycopeptide antibiotic family and has effective anti-tumor activity. Its main toxic side effect is pulmonary fibrosis. In order to further investigate the efficacy and mechanism of β-Nicotinamide Mononucleotide, researchers conducted further experiments. Administer BLM to mice through cough method and start administering NMN at a dose of 500 mg/kg/day. After 21 days of gastric lavage, age-related indicators were detected.

The results showed that the appearance of the lungs in the β-Nicotinamide Mononucleotide group was significantly improved, and the lung weight was significantly reduced. Through HE staining of mouse lung tissue, we found that NMN can alleviate alveolar destruction and interstitial inflammatory infiltration caused by BLM.

In addition, in flow cytometry of lung tissue and alveolar lavage fluid, researchers found a significant reduction in inflammatory cells in the group with added β-Nicotinamide Mononucleotide. And the immunohistochemical staining results showed that the expression of age-related cycle inhibitor proteins was significantly reduced in the NMN supplemented group.

The findings of these studies indicate that long-term addition of β-Nicotinamide Mononucleotide can effectively improve in vivo and in vitro ACE replication and stress-induced aging. Supplementing NMN may be a new and effective method for preventing and reducing age-related lung diseases and stress induced lung injury in the future.

It should be noted that ACEs have a high level of pro-inflammatory cytokine phenotype, but this age-related phenotype has not been detected in the study. In addition, there is currently relatively limited research data, so further in-depth exploration of the more comprehensive impact of β-Nicotinamide Mononucleotide on lung health is needed.
