NMN reduces virus-mediated inflammation

Recently, Scientific Reports published a research paper titled "Nicotinamide Mononucleotide (NMN) alleviates the poly(I:C)-induced inflammatory response in human primary cell cultures" online. The study found that NMN can reduce virus-mediated inflammation and help protect against viruses.


The researchers confirmed through RT-PCR that the expression levels of IL-6 and IL-12 mRNA in lung cells after simulated virus infection were significantly increased, indicating that the inflammatory response of lung cells was successfully induced. To evaluate the effects of high-dose NMN in cell samples induced or uninduced by viral infection, we also determined changes in characteristic gene expression levels of all modules between control and high-dose NMN.


Regarding the regulatory mechanism of the inflammatory response induced by NMN treatment, viral infection-induced activation reduced the expression level of TNFRSF10D, while the NMN group increased the expression level of this gene. In addition, the expression of genes related to translation and viral infection/transcription pathways was reduced due to induction of viral infection, indicating that NMN plays an important role in the restoration of inflammation-related gene expression levels. Further analysis showed that NMN treatment triggered the viral infection/transcription pathway, and this gene pathway had a direct causal relationship with the reduced expression levels of gene groups including IL6. In addition, this study found that under conditions of induced activation, NMN treatment may trigger an increase in the expression levels of genes related to the platelet-derived growth factor binding pathway and reduce the inflammatory response.
