The mechanism of SAMe

 

SAMe is an important physiologically active substance in the human body and has a wide range of functions, including metal detoxification and catecholamine metabolism, membrane fluidity, gene expression, and cell growth, differentiation, and apoptosis. As an important metabolic pleiotropic molecule, the important basis for its function is the transmethylation, transsulfation and propylamino effects of the methyl group, thio group and propylamino group in the SAMe structure in the metabolic pathway.

SAMe transmethylation can reduce the cholesterol/phospholipid ratio of liver cell membranes and red blood cell membranes, improve the fluidity of liver cell membranes, and increase the activity of Na+/K+-ATPase on the cell membrane to improve bile secretion and flow dynamics. Transsulfation increases glutathione levels and improves liver cells' ability to resist free radicals and detoxify; at the same time, glutathione transporter can transport glutathione into bile without relying on bile salts to stimulate bile flow. Transpropylation promotes normal liver cell regeneration and damaged liver cell repair, and inhibits inflammatory cytokines.

Based on the above three major mechanisms of action (transmethylation, transsulfation and transpropylamino), SAMe plays an important role in patients with hepatocellular cholestatic liver diseases, such as alcoholic liver disease, non-alcoholic fatty liver disease, and viral hepatitis. , drug-induced cholestasis and intrahepatic cholestasis of pregnancy.
