What are the potential effects of NMN on the human body?

NAD+ is an essential cofactor for key cellular physiological activities such as cell metabolism, cell death, aging, DNA repair, gene expression, and neuroinflammation. The concentration of NAD+ decreases with age in tissues such as the skin, blood, liver, muscle, and brain. It has been proven that supplementing with NAD+ precursors such as NMN and nicotinamide riboside (NR) can increase the concentration of NAD+ in the blood and tissues of mice, and may help reduce age-related diseases such as oxidative stress, DNA damage, neurodegeneration, and inflammatory responses.

Some articles have reviewed published studies on NMN in human clinical trials, which have assessed the safety, tolerability, and effects on NAD+ levels and related metabolic biomarkers of NMN.


1. Anti-aging effects: NMN supplementation has a positive impact on muscle strength and performance in the elderly, such as improving walking speed, grip strength, and the frequency of the 30-second chair stand test. In addition, NMN supplementation is also associated with potential anti-aging effects such as improved sleep quality, reduced blood pressure and blood sugar levels, and increased telomere length.


2. Changes in NAD+ levels: NMN supplementation can significantly increase the concentration of NAD+ and its metabolites in the blood. For example, one study found that taking 250 mg of NMN orally every day for 12 weeks could significantly increase the concentration of NAD+ in the whole blood of healthy participants.

3. Insulin sensitivity in women with prediabetes: In a study involving 25 postmenopausal women with prediabetes, NMN supplementation significantly improved muscle insulin sensitivity and increased the expression of genes related to muscle remodeling.


4. Safety: In a trial conducted in Japan, no significant clinical symptoms or harmful effects were observed after 10 healthy men took a single oral dose of NMN (up to 500 mg). In another trial conducted in the United States, 25 postmenopausal women with prediabetes took 250 mg of NMN orally every day for 10 weeks. The results showed that NMN increased muscle insulin sensitivity, but no effects on skeletal muscle NAD+ concentration were observed.
