NMN Supplementation Improves Muscle Strength in Older Healthy Men
Preclinical studies suggest that elevations in nicotinamide adenine dinucleotide (NAD+) following administration of the NAD++ precursor Nicotinamide Mononucleotide (NMN) can alleviate aging-related diseases; however, this Human data is limited.
Recent studies have found that long-term oral NMN increases blood NAD+ levels and alters physiological dysfunction in healthy older participants.
In a placebo-controlled, randomized, double-blind, parallel-group trial, researchers gave older men 250 mg of NMN daily for 6 or 12 weeks.
Long-term supplementation with NMN was well tolerated and caused no apparent deleterious effects.
Metabolomic analysis of whole blood samples showed that oral NMN supplementation significantly increased NAD+ and NAD+ metabolite concentrations.
There were nominally significant improvements in gait speed and performance on the left grip strength test, which should be validated in larger studies;
however, NMN had no significant effect on body composition.
Therefore, long-term oral NMN supplementation can be an effective NAD+ booster for preventing aging-related muscle dysfunction in humans.
Chronic oral NMN administration increases whole blood NAD+ and NAD+ related metabolite levels.

Notably, NMN also significantly elevated the levels of nicotinic acid mononucleotide (NAMN) (an intermediate of the NAD+ de novo synthesis pathway) and nicotinic acid riboside (NAR).
Effect of NMN on muscle mass.

NMN partly improves muscle strength and performance.

In this study, long-term oral supplementation with 250 mg of NMN per day was safe and a well-tolerated and effective strategy for promoting NAD+ metabolism in healthy older men.
In addition, exploratory analyses of the effects of NMN supplementation on physiological function suggest that NMN can improve muscle strength, an important clinical indicator of aging.
