Supplementation With NMN May Improve Lung Damage Caused By COVID-19

Recently, a team from the Chinese Academy of Sciences, together with Peking University and Tsinghua University, published their preclinical research in the top journal "Cell Discovery".

They found that NMN supplementation has a similar NAD+ effect on the new crown.

It can not only improve the NAD+ metabolism caused by infection with the new crown, but also It can also improve the dysregulation of immune response and cell death-related gene expression, and inhibit cell death.

To examine whether NAD+ intermediates have a similar therapeutic effect on pneumonia caused by SARS-CoV-2, the researchers used a more severe infection model developed recently in which mice were infected with a mouse-adapted SARS-CoV-2 MASCp36 12.

Infected mice (8-9 months old) were treated with NMN or NAD+.

Doses of NMN are widely used in mouse models and are equivalent to 55 mg/kg/day in humans based on body surface area.

Compared with the severe inflammatory cell infiltration and alveolar septal thickening detected in the saline group, pathological damage was significantly reduced in both the NMN and NAD+ groups.

Even more strikingly, 30% of the mice ended up surviving in the NMN group, while 100% of the mice in the saline group died at 8 dpi.

In addition, the number of CD68+ cells and the expression of Cd200r3 were significantly decreased in both NMN and NAD+ groups.

Furthermore, NMN and NAD+ significantly inhibited cell death.

The study showed that NMN supplementation rescued global changes in TCA circulating metabolites, and NAD+, NMN, NR, and NAM were elevated in the lungs of infected mice after NMN supplementation.

Targeted metabolomic analysis of the study showed that metabolic disturbances in the sera of COVID-19 patients were partially reproduced in the lungs of mouse models and could be ameliorated by NMN supplementation.
