Aging and obesity are major risk factors for various diseases. Studies have proven that NAD+ levels will decrease with aging and obesity. Nicotinamide Mononucleotide (NMN) is a bioactive nucleotide that can be synthesized naturally and is the precursor of NAD+. The body can increase NAD+ levels in the body through dietary supplements and reduce factors related to aging and obesity.

Recently, a Japanese research team conducted an 8-week, single-center, single-arm, open-label clinical trial. 11 healthy middle-aged Japanese men took 125 mg NMN capsules daily before breakfast. The study proved that 8-week oral administration of Safety of NMN, and NAD+ levels in peripheral blood mononuclear cells (PBMCs) increased with NMN administration.

In addition to this, another result of this study showed the effect of NMN on glucose metabolism, 8 weeks of NMN intake moderately attenuated the effect of postprandial hyperinsulinemia, suggesting that NMN may moderately alleviate the involvement of postprandial insulin resistance the possibility of hyperinsulinemia in patients.

Of course, the sample size of this trial is small and has limitations. We look forward to subsequent more comprehensive studies on the impact of NMN in the treatment of hyperinsulinemia. The potential of NMN deserves further exploration.

Overall, this trial demonstrates that NMN at 250 mg/d for 8 weeks is safe and increases PBMC NAD+ levels.
